Large Scale Production and Purification of recombinant Staphylokinase, at the Biochemical Engineering Research and Process Development Centre, (BERPDC), Institute of Microbial Technology (IMTECH), Chandigarh. (2002-2007).
The technology developed & transferred successfully to Strides Acrolabs, Bangalore.
The process was undertaken at a production level of 90 mgL-1 staphylokinase, at shake flask.
Classical and statistical optimization methods used for optimization of fermentation medium.
Production of Staphylokinase was increased to 450 mgL-1 at 20 L fermenter level after first round of optimization of medium and process parameters like inducer concentration, induction time, MIC, selection pressure, plasmid stability etc.
Production taken to 3600 mgL-1 (40x) after optimization of fed-batch fermentation, feed rate, feed composition etc.
Cell lysis was optimized at large scale using bead milling, for critical parameters, with recovery of more than 90 % of Staphylokinase.
Purification was optimized in a two-step process initially with more than 50 % recovery using ion-exchange chromatography and hydrophobic interaction chromatography.
pH based chromatography method was innovated and optimized with recovery more than 70 % and with more than 95 % homogeneity.
Other Projects (JNU-CSIR)
Evaluation of WIDAL test in the detection of Antibodies against Salmonella typhi, in suspected enteric fever cases, in the Dept. Of Microbiology, AIIMS, New Delhi, (Jan – Feb 02). The authenticity of the test and its accuracy was estimated in patients registering in AIIMS for suspected typhoid in Jan-Feb 02. After the six weeks study it was found that the WIDAL test gave false positive results in most of the cases. And it should not be considered a diagnostic test for typhoid fever.
